P-132 Underrepresentation of racial and ethnic minorities in metastatic colorectal carcinoma clinical trials within the United States

Representation of vulnerable subpopulations is essential for generalizability when designing clinical trials. This study aims to investigate if underrepresentation racial and ethnic minorities exists in metastatic colorectal carcinoma (CRC) trials the United States. The ClinicalTrials.gov database was queried within States with start date 2004-2020. Incidence data patients extracted from SEER Research Plus Database (17 registries, 2000-2019) divided into 5-year subgroups (2000-2004, 2005-2009, 2010-2014, 2015-2019). Enrollment fraction (EF) defined as number trial participants by US incidence CRC each race, ethnicity, gender. ratio (ER) proportion subgroup subgroup. Wilcoxon rank sum tests were employed determine distribution ERs. 8555 pooled 86 analyzed database. 49 (57.6%) had locations Northeast, 54 (63.5%) South, 37 (43.5%) Midwest, 40 (47.1%) West. Only 51.2% reported race data, 32.6% ethnicity 100% Of white represented 79.4%, Black 5.0%, Asian/Pacific Islander (API) 13.0%, American Indian/Alaska Native (AIAN) 0.1%. non-Hispanic 93.1%, while Hispanic 5.6% patients. (median ER 0.36, p 0.05). Clinical geographic location did not affect EF or (p>0.05). Strategies are needed increase enrollment Black, API, AI/IN, carcinoma. Identification systemic barriers preventing these will be integral public policy advocacy representation.
